Target Description
Acetylcholinesterase (AChE) is one of the most important enzymes involved in nerve transmission. The enzyme is bound to cellular membranes of excitable tissue (synaptic junction, endoplasmic reticulum, etc.). Acute toxicity to humans and animals through inhibition of AChE by both nerve gases and an important class of pesticides has long been a field of intensive scientific investigation. AChE inhibitors have also been used clinically as Alzheimer’s treatments (e.g., tacrine (tetrahydroaminoacridine)) and are the subject of increasing interest in various disease processes and treatment strategies. However, both environmental detection of AChE inhibitors and development of modulators of AChE enzymatic activity as drugs have been hampered by the difficulty and complexity of the current assay methods. We have developed a highly sensitive, very rapid, extremely simple assay for AChE activity, using the natural substrate, acetylcholine. As shown in Figure 1, a series of coupled enzyme reactions quickly translates the presence of active AChE into a change in the luminance of the reaction. First (reaction I), acetylcholine is hydrolyzed by the AChE to yield acetate and choline. The acetate and choline then enter a coupled enzyme reaction (reaction II) that results in consumption of ATP, and finally the ATP concentration is measured by the Well-established luciferase method (reaction III). These reactions can occur simultaneously, and the result is generally obtained in five minutes or less. Inhibitors of AChE are readily detected by an increase in luminance due to reduced consumption of ATP.

Frequently Asked Questions

What is the catalog number for Bioluminescent Acetylcholinesterase Assay Kit?

The catalog number for Bioluminescent Acetylcholinesterase Assay Kit is CLACHE100-3.

Is Bioluminescent Acetylcholinesterase Assay Kit in stock?

Bioluminescent Acetylcholinesterase Assay Kit is currently made to order / not held in immediate stock. Contact us for lead time.

What is Bioluminescent Acetylcholinesterase Assay Kit used for?

A rapid and extremely sensitive assay for acetylcholinesterase using the natural substrate acetylcholine. A series of coupled reactions generate a bioluminescent signal. Ideal for identifying and characterizing acetylcholinesterase inhibitors.

What temperature does this product ship at?

Shipping temperature is chosen based on the product’s stability needs, ranging from ambient to dry ice, and is indicated on the product page. Any related shipping fee is shown clearly in your quotation.

What is the molecular weight of the target protein?

Molecular weight information, where applicable, is listed on the datasheet under target details, useful for confirming the correct band on western blot.

How long will it take to receive this product if it’s custom made?

Lead times depend on the product type: custom polyclonal antibodies typically take 12-14 weeks, monoclonal antibodies 15-17 weeks, made-to-order recombinant proteins 4-8 weeks, and made-to-order recombinant antibodies 6-8 weeks. These are estimates and may shift due to production delays.
